I have a server and I have a emailing software. I want to send mass/bulk email to my customers list.(There is around 100K people, which I was trying gather this list for 10 years) and now I want to send them email.
Can I send mass email without using SMTP?

There are a couple of ways to send email without the need of your own SMTP server. There is an addition for PHP you can get called PHPMailer which will simplify the process. Or, you can send it with php's built in mail() function, but you must be sure to to format the header information correctly.

However, sending mail without using a verified SMTP server will most likely result in your email being sent to customers junk mail folder. If any emails are destined to corporate addresses, the corporations firewall may drop the email entirely. Typically if an email provider detects enough emails of this kind, it may even blacklist your IP / server for a time.
I dont understand how you tagged your post classic-asp and php. Both will run on IIS, but to send mail you will need to either run an SMTP server or use a SMTP service. If you want to save yourself a lot of heart-ache of running an SMTP server specially when it comes to handling bounced, unsubscribes, feedback loops and the such then you are much better off using a 3rd party service.
Services like Mandrill, MailGun, Amazon SES and others will allow you to send 100k messages a month on the cheap, without too much worry about keeping SMTP services running, ip-whitelists, and security ports. The catch is, if you use these services to SPAM you will be kicked off pretty quickly. This is why many of the services have warm up periods while you ramp up the number of messages you send. THis is different than if you used Outlook.com or Gmail smtp servers which with 100k messages you would surely get placed in the spam folder.
You may also want to checkout https://sendy.co/Its software you install on your server, but designed to work with Amazon SES. You create the messages using sendy.co but amazon does the delivery. Requires an Amazon AWS account, but its pretty nice as far as an admin. It also plays nice with existing code.
